A peptide antigen mimicking mycobacterial proteins was used and tested for the Cell Mediated Immune (CMI) response. All BCG strains and most non-tuberculous mycobacteria do not contain ESAT-6, CFP-10 and TB7.7, except for M. kansasii, M. szulgai and M. marinum. In infected patients, the lymphocytes can usually recognize that the antigen is from a tuberculosis complex organism or other mycobacteria. The identification process produces and secretes cytokines, namely INF-γ.
The principle of adaptive immunity produced by the human body against tuberculosis antigens can be used to diagnose latent infections and active diseases, independent of BCG and non-tuberculous mycobacteria, due to the use of M. tuberculosis specific antigen ESAT -6 and CFP-10 to stimulate the T cells of the patient's whole blood to see if they produce INF-γ. If INF-γ is produced, it means that the tuberculosis infection has been produced, and it can also be used to assist the diagnosis of active tuberculosis patients.
